What is NMN(CAS # 1094-61-7):A key NAD+ intermediate
Nicotinamide mononucleotide (NMN) is a type of bioactive nucleotide which is naturally formed by
the reaction between a phosphate group and a nucleoside containing ribose and nicotinamide. It
is a nucleotide that is most recognized for its role as an intermediate of Nicotinamide denine
dinucleotide (NAD+) biosynthesis.
Beneficial health effects:
• Anti-aging and longevity effect
• Mitochondrial health: rejuvenate mitochondria
• Better glucose metabolism
• Enhancing muscle regeneration
• Improving brain function and cardiovascular health
NMN or Nicotinamide Riboside(NR): Which One is Better
Among the diverse type of NAD+ precursors, until now only NMN and NR presented better
pharmacokinetic and pharmacological property. Accordingly, these two intermediates are now
widely utilized for clinical trials. Still, the question remains: Which one is better?
1. NR is approved GRAS in US, while NMN is not yet to prove the safety of human consumption
2. In the salvage pathway, Nicotinamide (NAM) and NR are firstly converted into NMN, and finally
turned NAD+, therefore, NMN is more accurate to be recognized as an intermediate of NAD+
and is more efficient to produce NAD+.
3. Additionally, NMN has quite a few strong advantages of its own:
• Friedreich’s Ataxia (FRDA), a rare inherited childhood heart disease, NMN was found
successful where NR treatment failed
• In case of treatment of cognitive impairment in Alzheimer’s disease, for which β amyloid
plaque is thought of as the main culprit, NMN was potent enough to reduce the burden of
amyloid β plaque by reducing its production
Why not consume NAD+ directly?
Due to the large molecule, NAD+ is hard to penetrate the cell membrance and then enter the cell
interior which NMN can through reasily, and two of NMN molecules will be combined
spontaneously for form an NAD+ molecule once entered the cell interior.
